These single-stranded regions have all been studied in transcriptionally active promoters. Whether the single-stranded DNA binding factors also continue to bind to the transcriptionally inactive promoters and, if so, what changes take place during their transition into a transcriptionally active state, need to be studied. In addition, mechanisms involved in generating isolated single-stranded sites at specific points within a predominantly double-stranded genome need to be examined in detail.
Proteins were then transferred to nitrocellulose membranes (Schleicher & Schuell, Keene, NH, USA). These were saturated by incubation at 4 °C overnight with 10% non-fat dry milk in NaCl/Pi/0. 1% Tween 20 and then incubated with the rabbit polyclonal anti-[single-stranded D-box-binding factor (ssDBF)] serum for 60 min at room temperature. The rabbit polyclonal anti-ssDBF serum was used at a dilution of 1 : 1000 in NaCl/Pi/0.
