Metachromatic leukodystrophy : Deficiency of Aryl sulphatase A or Sphingolipid activator protein (Saponin-B)= accumulation of lipid sulphatide in the myelin sheath, brain and peripheral nerve. Cognitive function is minimally affected initially followed by motor difficulties and dysarthria. Diagnosis is by estimating Aryl sulphatase A in leukocytes or fibroblasts.
