Differentially Protected Orthogonal Lanthionine Technology (DPOLT) for solid phase synthesis of peptides is disclosed herein. The technology depends on the bulk manufacture of various orthogonally protected peptide bridges whose active carboxyl and amino protecting groups can be differentially removed. The orthogonally protected peptide bridges can be used in, for example, solid phase peptide synthesis, to prepare conformationally constrained bioactive peptides containing intramolecular bridges forming ring structures.
